چکیده

Electric field-induced locomotion and transport of worms Caenorhabditis elegans (C. elegans) in a microfluidic device (electrotaxis) is demonstrated in this paper. Direct (DC) and alternating (AC) electric fields inside a microchannel were used to stimulate worms locomotion towards the negative pole and to halt their locomotion while localizing them at desired locations in the channel respectively. This technique promises to facilitate the study of movement-related disorders (e.g., Parkinson’s Disease) and to perform drug discovery assays on a chip. We also demonstrate an application of our system in separating worms based on their age and size from a mixed culture.

Electrical sorting of Caenorhabditis elegans.

The nematode (worm) C. elegans is one of the widely studied animal model organisms in biology. It develops through 4 larval stages (L1-L4) in 2 to 3 days before becoming a young adult. Biological assays involving C. elegans frequently require a large number of animals that are appropriately staged and exhibit a similar behaviour. We have developed a new method to synchronize animals that relies...

Advanced Behavioral Analyses Show that the Presence of Food Causes Subtle Changes in C. elegans Movement

As a widely used and studied model organism, Caenorhabditis elegans worms offer the ability to investigate implications of behavioral change. Although, investigation of C. elegans behavioral traits has been shown, analysis is often narrowed down to measurements based off a single point, and thus cannot pick up on subtle behavioral and morphological changes. In the present study videos were capt...

Using Caenorhabditis elegans as a model system to study protein homeostasis in a multicellular organism.

The folding and assembly of proteins is essential for protein function, the long-term health of the cell, and longevity of the organism. Historically, the function and regulation of protein folding was studied in vitro, in isolated tissue culture cells and in unicellular organisms. Recent studies have uncovered links between protein homeostasis (proteostasis), metabolism, development, aging, an...

A sorting strategy for C. elegans based on size-dependent motility and electrotaxis in a micro-structured channel.

Caenorhabditis elegans (C. elegans) is a model organism widely utilized in various fundamental studies in developmental, neural and behavioural biology. The worm features four distinct larval stages, and many research questions are stage-specific; therefore, it is necessary to sort worms by their developmental stages, which are typically represented by different size ranges. However, manually s...

Analysis of the roles of kinesin and dynein motors in microtubule-based transport in the Caenorhabditis elegans nervous system.

The heteromeric kinesins constitute a subfamily of kinesin-related motor complexes that function in several distinct intracellular transport events. The founding member of this subfamily, heterotrimeric kinesin II, has been purified and characterized from early sea urchin embryos, where it was shown using antibody perturbation to be required for the synthesis of motile cilia, presumably by driv...
